(1). 抗原或抗體的固相化及抗原或抗體的酶標記。
(2). 結(jié)合在固相載體表面的抗原或抗體仍保持其免疫學活性。
(3). 酶標記的抗原或抗體既保留其免疫學活性，又保留酶的活性。
(4). 受檢標本與固相載體表面的抗原或抗體起反應(yīng)。再加入酶標記的抗原或抗體，也通過反應(yīng)而結(jié)合在固相載體上。
(5). 此時固相上的酶量與標本中受檢物質(zhì)的量呈一定的比例。
(6). 加入酶反應(yīng)的底物后，底物被酶催化成為有色產(chǎn)物，產(chǎn)物的量與標本中受檢物質(zhì)的量直接相關(guān)，故可根據(jù)呈色的深淺進行定性或定量分析。測定方法具有很高的敏感度（pg-ng/ml水平），并且重復(fù)性好。以免疫學反應(yīng)為基礎(chǔ)，將抗原、抗體的特異性反應(yīng)與酶對底物的高效催化作用相結(jié)合起來的一種敏感性很高的試驗技術(shù)。

保存條件  Store at -20 °C for one year. Avoid repeated freeze/thaw cycles. The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20°C. When reconstituted in sterile pH 7.4 0.01M PBS or diluent of antibody the antibody is stable for at least two weeks at 2-4 °C.
