封閉血清適用于免疫組化或者免疫熒光實(shí)驗(yàn)中樣本的封閉??梢苑忾]待檢樣本中與蛋白質(zhì)非特異性結(jié)合的位點(diǎn),另外還可以封閉樣品中內(nèi)源性的Fc片段結(jié)合位點(diǎn),阻斷抗體與樣品中的Fc受體結(jié)合,從而降低背景,減少假陽性。在選擇封閉血清時(shí),要注意封閉用血清中不能含有目標(biāo)蛋白,且與一抗來源不同,可用與二抗相同來源的封閉血清。
封閉兔血清(原液)使用說明書使用方法:
使用時(shí),用PBS(0.01mol/LpH7.2-7.4)將封閉血清原液稀釋10或20倍,配成10%或者5%的工作液,直接滴加在組織切片或者細(xì)胞凃片上,37℃孵育10-30分鐘,清除血清,勿洗,滴加適當(dāng)比例稀釋的一抗工作液,37℃孵育1-2h或4℃過夜。

as a trans-activator that acts to negatively regulate cell division by controlling a set of genes required for this process. One of the activated genes is an inhibitor of cyclin-dependent kinases. In vitro, the interaction of TP53 with cancer-associated/HPV (E6) viral proteins leads to ubiquitination and degradation of TP53 giving a possible model for cell growth regulation.
