Western Blot Marker是專(zhuān)門(mén)為Western Blot試驗(yàn)設(shè)計(jì)的分子量標(biāo)準(zhǔn)。傳統(tǒng)的Western Blot試驗(yàn)需要使用預(yù)染蛋白Marker來(lái)跟蹤檢測(cè)陽(yáng)性抗原信號(hào)和轉(zhuǎn)膜效率。但預(yù)染蛋白Marker用在Western Blot試驗(yàn)中有三大缺點(diǎn):(1)預(yù)染蛋白Marker無(wú)法在膠片上曝光,曝光信號(hào)需要根據(jù)膜上的預(yù)染蛋白Marker來(lái)判斷,因此后續(xù)的圖片需要拼接才能兩者合而為一;(2)預(yù)染蛋白Marker都經(jīng)過(guò)化學(xué)修飾,經(jīng)過(guò)化學(xué)修飾的蛋白在SDS-PAGE電泳過(guò)程中遷移率發(fā)生改變,分子量信息不再準(zhǔn)確,往往導(dǎo)致Western Blot結(jié)果判斷出現(xiàn)較大偏差;(3)預(yù)染蛋白Marker無(wú)法檢測(cè)作為雜交過(guò)程的陽(yáng)性對(duì)照。Western Blot Marker由6種不同分子量的蛋白組成(20、40、60、70、80、120 KDa),其中70KDa條帶為藍(lán)色預(yù)染marker,其它五個(gè)條帶都可與人、小鼠、大鼠、山羊、綿羊、兔的IgG恒定區(qū)結(jié)合。Western Blot Marker可以直接結(jié)合二抗,或通過(guò)結(jié)合一抗間接結(jié)合二抗,因此蛋白Marker就能與抗原在同一張膠片上曝出信號(hào),陽(yáng)性信號(hào)可以直接通過(guò)Western Blot Marker的位置來(lái)判斷。Western Blot Marker不經(jīng)過(guò)任何化學(xué)修飾,分子量精確。

Function : Tumor suppressor. Acts as a dual-specificity protein phosphatase, dephosphorylating tyrosine-, serine- and threonine-phosphorylated proteins. The lipid phosphatase activity is critical for its tumor suppressor function. Antagonizes the PI3K-AKT/PKB signaling pathway by dephosphorylating phosphoinositides and thereby modulating cell cycle progression and cell survival. The unphosphorylated form cooperates with AIP1 to suppress AKT1 activation. Dephosphorylates tyrosine-phosphorylated focal adhesion kinase and inhibits cell migration and integrin-mediated cell spreading and focal adhesion formation. Plays a role as a key modulator of the AKT-mTOR signaling pathway controlling the tempo of the process of newborn neurons integration during adult neurogenesis, including correct neuron positioning, dendritic development and synapse formation. May be a negative regulator of insulin signaling and glucose metabolism in adipose tissue. The nuclear monoubiquitinated form possesses greater apoptotic potential, whereas the cytoplasmic nonubiquitinated form induces less tumor suppressive ability.
