活性氧(Reactive oxygen species, ROS) 包括超氧自由基、過(guò)氧化氫、及其下游產(chǎn)物過(guò)氧化物和羥化物(O2•−,H2O2, •OH, ONOO−, •NO)等,參與細(xì)胞生長(zhǎng)增殖、發(fā)育分化、衰老和凋亡以及許多生理和病理過(guò)程。采用2,7-dichlorofluorescin diacetate (DCFH-DA)是迄今Z常用、Z靈敏的細(xì)胞內(nèi)活性氧檢探針。DCFH-DA 沒(méi)有熒光,進(jìn)入細(xì)胞后被酯酶水解為dichlorofluorescin (DCFH)。在活性氧存在時(shí)DCFH 被氧化為不能透過(guò)細(xì)胞膜的強(qiáng)綠色熒光物質(zhì)dichlorofluorescein (DCF),其熒光在激發(fā)波長(zhǎng)502 nm,發(fā)射波長(zhǎng)530 nm 附近有Z大波峰,強(qiáng)度與細(xì)胞內(nèi)活性氧水平成正比。此活性氧檢測(cè)系統(tǒng)本底低,靈敏度高,重復(fù)性好,操作簡(jiǎn)便。
本試劑盒適于熒光顯微鏡、激光共聚焦顯微鏡等圖像檢測(cè),熒光分光光度計(jì)、熒光酶標(biāo)儀、流式細(xì)胞儀定量ROS 檢測(cè)。

The gene product catalyzes the formation of all trans-polyprenyl pyrophosphates from isopentyl diphosphate in the assembly of polyisoprenoid side chains, the first step in coenzyme Q biosynthesis. Defects in this gene are a cause of coenzyme Q10 deficiency.
