(1). 抗原或抗體的固相化及抗原或抗體的酶標(biāo)記。
(2). 結(jié)合在固相載體表面的抗原或抗體仍保持其免疫學(xué)活性。
(3). 酶標(biāo)記的抗原或抗體既保留其免疫學(xué)活性,又保留酶的活性。
(4). 受檢標(biāo)本與固相載體表面的抗原或抗體起反應(yīng)。再加入酶標(biāo)記的抗原或抗體,也通過反應(yīng)而結(jié)合在固相載體上。
(5). 此時固相上的酶量與標(biāo)本中受檢物質(zhì)的量呈一定的比例。
(6). 加入酶反應(yīng)的底物后,底物被酶催化成為有色產(chǎn)物,產(chǎn)物的量與標(biāo)本中受檢物質(zhì)的量直接相關(guān),故可根據(jù)呈色的深淺進行定性或定量分析。測定方法具有很高的敏感度(pg-ng/ml水平),并且重復(fù)性好。以免疫學(xué)反應(yīng)為基礎(chǔ),將抗原、抗體的特異性反應(yīng)與酶對底物的高效催化作用相結(jié)合起來的一種敏感性很高的試驗技術(shù)。
