背景介紹：SPFH2 is a ubiquitously expressed 339 amino acid protein that belongs to the band 7/mec-2 family. Localized to lipid raft-like domains in the membrane of the endoplasmic reticulum (ER), SPFH2 plays a crucial role in the ER-associated degradation (ERAD) pathway that removes metabolically regulated and aberrant proteins from the ER. Specifically, SPFH2 associates with IP3R-I (Inositol 1,4,5-triphosphate receptor I), a substrate of the ERAD pathway, and facilitates its polyubiquitination and subsequent degradation. Three isoforms of SPFH2 are expressed due to alternative splicing events.

組織中非抗原抗體反應(yīng)出現(xiàn)的陽(yáng)性染色稱為非特異性背景染色，zui常見(jiàn)的原因是蛋白吸附于高電荷的膠原和結(jié)締組織成分上。zui有效方法是在用*抗體前加與制備第二抗體相同動(dòng)物的非免疫血清（1:5-1:20），封閉組織上帶電荷基團(tuán)而除去與一抗非特異性結(jié)合。必要時(shí)加入2%-5%牛血清白蛋白，可進(jìn)一步減少非特異性染色，作用時(shí)間為10-20min。免疫熒光染色時(shí)，可用0.01%伊文藍(lán)（PBS溶液）稀釋熒光抗體，對(duì)消除背景的非特異性熒光染色有很好的效果。洗滌用的緩沖液中加入0.85%-1%NaCl成為高鹽溶液，充分洗滌切片，能有效的減少非特異性結(jié)合而減少背景染色。
