背景介紹：The protein encoded by this gene is a cell-surface glycoprotein found on melanoma cells. The protein shares sequence similarity and iron-binding properties with members of the transferrin superfamily. The importance of the iron binding function has not yet been identified. This gene resides in the same region of chromosome 3 as members of the transferrin superfamily. Alternative splicing results in two transcript variants.

流式檢測樣品的制備

提示

1、流式細胞儀檢測需要將細胞制備成單細胞懸液。

2、避免細胞成團，以免堵塞流式細胞儀。

3、實體組織制備單細胞懸液需要物理方法分離或酶消化法進行，具體組織的方法，依照經(jīng)驗來進行。

方案A: 培養(yǎng)細胞的制備

實驗材料：0.25%胰，15ml或50ml的離心管。

實驗流程：

1、如果是懸浮細胞，則小心將細胞移入離心管中，進行計數(shù)和活力分析。進行步驟3。

2、如果是貼壁細胞，建議使用0.25%的*消化細胞，制備成單細胞懸液后置于離心管中計數(shù)和活力分析。

3、200g 離心5min。棄去上清，應(yīng)用適量的流式孵育緩沖液重懸細胞，調(diào)整細胞濃度為1×107/ml。

方案B: 組織細胞制備。

實驗材料與試劑：剪刀和鑷子，載玻片， 60×15mm的組織培養(yǎng)皿；細胞過濾器（尼龍網(wǎng)過濾）,0.01MPBS。
