背景介紹：The protein encoded by this gene was first identified by its binding to the APC protein which is often mutated in familial and sporadic forms of colorectal cancer. This protein localizes to microtubules, especially the growing ends, in interphase cells. During mitosis, the protein is associated with the centrosomes and spindle microtubules. The protein also associates with components of the dynactin complex and the intermediate chain of cytoplasmic dynein. Because of these associations, it is thought that this protein is involved in the regulation of microtubule structures and chromosome stability. This gene is a member of the RP/EB family.

關(guān)于肽鏈的設(shè)計

多肽是大分子，每條多肽序列在物理和化學(xué)特性上都有自己的*性。有些多肽合成起來很困難，還有些合成相對容易，但純化困難，其主要是不溶于水，所以在純化中，那些疏水肽必須溶于非水溶劑中或特殊的緩沖液，然而這些溶劑或緩沖液可能不適合應(yīng)用于生物實驗系統(tǒng)，研究人員不能使用該多肽達到研究目的。

一、變難為易

1、減少序列長度

肽的長度增加導(dǎo)致粗產(chǎn)物純度降低，小于15個殘基的肽較容易得到.當肽鏈增加到20個以上時，正常產(chǎn)物的量就要考慮。在具體實踐中殘基低于20往往能得到更好的結(jié)果；

2、減少疏水殘基

疏水殘基占明顯優(yōu)勢的肽，尤其在距C端7―12個殘基的區(qū)域，會引起合成困難，一般認為由于合成中形成β折疊片，而產(chǎn)生不*配對。當然，可用幾個極性殘基置換，或加入Gly或Pro易打開肽結(jié)構(gòu)可能會得到幫助；

3、減少“難度”殘基

有多個Cys、Met、Arg、Try殘基一般難以合成，Ser通?？勺鳛榉茄趸鎿Q。
