背景介紹：NHN1 is a 953 amino acid nuclear protein that contains one C3H1-type zinc finger and exists as two alternatively spliced isoforms. The gene that encodes NHN1 contains more than 61,500 bases and maps to human chromosome 16q24.2. Encoding over 900 genes and consisting of approximay 90 million base pairs, chromosome 16 makes up nearly 3% of the human genome and is associated with a variety of genetic disorders. The GAN gene is located on chromosome 16 and, when mutated, may lead to giant axonal neuropathy, a nervous system disorder characterized by increasing malfunction with growth. Alterations in the CREB gene and NOD2 gene, both of which are located on chromosome 16, results in Rubinstein-Taybi syndrome and Crohn's disease, respectively. An association with systemic lupus erythematosis and a number of other autoimmune disorders with the pericentromeric region of chromosome 16 has led to the identification of SLC5A11 as a potential autoimmune modifier.

福爾馬林固定的組織有可能破壞了原來(lái)組織的抗原結(jié)構(gòu)，蛋白多肽發(fā)生交聯(lián)，導(dǎo)致部分抗原表位封閉，結(jié)合位點(diǎn)減少，所以需要抗原修復(fù)，以暴露原來(lái)的抗原表位，達(dá)到充分顯露抗原，以不出現(xiàn)明顯脫片現(xiàn)象為佳。

抗原修復(fù)的幾種常用方法（供參考）

1、 微波爐加熱：

在微波爐里加熱0.01*緩沖液（pH6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復(fù)1-2次。根據(jù)玻片情況可參考采用微波加熱“3-5-3法”3分鐘溫火沸騰后靜止5分鐘，再溫火沸騰3分鐘即可。

適用的抗原：來(lái)源于人、大鼠、小鼠的組織標(biāo)本；來(lái)源于其他動(dòng)物種屬的組織標(biāo)本。
