背景介紹：This gene encodes a zinc-dependent aminopeptidase that cleaves vasopressin, oxytocin, lys-bradykinin, met-enkephalin, dynorphin A and other peptide hormones. The protein can be secreted in maternal serum, reside in intracellular vesicles with the insulin-responsive glucose transporter GLUT4, or form a type II integral membrane glycoprotein. The protein catalyzes the final step in the conversion of angiotensinogen to angiotensin IV (AT4) and is also a receptor for AT4. Alternative splicing results in multiple transcript variants encoding different isoforms.

冰凍切片的免疫組化染色步驟

- 速凍組織

- 冰凍切片4~8μm，冰凍切片后如不染色，必須吹干，儲存低溫冰箱內(nèi)，或進(jìn)行短暫預(yù)固定后儲存于-20℃冰箱

- 室溫放置30分鐘后，入4℃丙酮固定10分鐘，也可根據(jù)需要選擇其他的固定方式

- PBS沖洗，5分鐘×3次

- 用3%過氧化氫孵育5~10分鐘，消除內(nèi)源性過氧化物酶的活性
