Anti-CK7抗體-細胞角蛋白7抗體產品介紹：Cytokeratins are a subfamily of intermediate filament proteins and are characterized by a remarkable biochemical diversity, represented in human epithelial tissues by at least 20 different polypeptides. They range in molecular weight between 40 kDa and 68 kDa and isoelectric pH between 4.9 – 7.8. The individual human cytokeratins are designated 1 to 20. The various epithelia in the human body usually express cytokeratins which are not only characteristic of the type of epithelium, but also related to the degree of maturation or differentiation within an epithelium. Cytokeratin subtype expression patterns are used to an increasing extent in the distinction of different types of epithelial malignancies. The cytokeratin antibodies are not only of assistance in the differential diagnosis of tumors using immunohistochemistry on tissue sections, but are also a useful tool in cytopathology and flow cytometric assays.

福爾馬林固定的組織有可能破壞了原來組織的抗原結構，蛋白多肽發(fā)生交聯(lián)，導致部分抗原表位封閉，結合位點減少，所以需要抗原修復，以暴露原來的抗原表位，達到充分顯露抗原，以不出現(xiàn)明顯脫片現(xiàn)象為佳。

抗原修復的幾種常用方法（供參考）
1、 微波爐加熱：
在微波爐里加熱0.01*緩沖液（pH6.0）至沸騰后將組織芯片放入，斷電，間隔5-10分鐘，反復1-2次。根據玻片情況可參考采用微波加熱“3-5-3法”3分鐘溫火沸騰后靜止5分鐘，再溫火沸騰3分鐘即可。
適用的抗原：來源于人、大鼠、小鼠的組織標本；來源于其他動物種屬的組織標本。
2、 沸熱修復：
電爐或水浴鍋加熱0.01M*緩沖液（pH6.0）至95℃左右，放入組織芯片加熱10-15分鐘。
3、 高壓熱修復：
在沸水中加入EDTA（pH8.0）或0.01m*緩沖溶液（pH6.0）.蓋上不銹鋼鍋蓋，但不能鎖定。將玻片置于金屬染色架上，緩慢加壓，使玻片在緩沖液中浸泡五分鐘，然后將蓋子鎖定，小閥門將會升起來。10分鐘后除去熱源，置入涼水中，當小閥門沉下去后打開蓋子。此方法適用于較難檢測或核抗原的抗原修復。（骨及軟骨組織的標本選擇較溫和的微波加熱修復） 。
4、 酶消化方法：
常用0.1%*和0.4%*液。*使用前預熱37℃，消化時間約為5-30分鐘。
適用于被固定遮蔽的抗原：包括 Collagen.GFAP. Complement.Cytokeratin.c-erB-2.LCA.LN.等。

抗體修復方法：
方法1：沸水浴修復，將盛有修復液和玻片的燒杯置于沸水浴環(huán)境，保持外部沸騰狀態(tài)15min，自然冷卻至室溫；
方法2：微波修復，將盛有修復液和玻片的燒杯置于微波爐中，高火5min，停火3min，中火5min，自然冷卻至室溫。
